Puncture rear tyre Stromer ST2 with carbon belt

I've got a very small puncture on the rear tyre of my ST2. When I'm trying to inflate this tyre, all the air goes out directly from the tire valve. I do not understand how this Pirelli tyres have got a tube.

I have absolutely no idea what you're asking. You start with a puncture in your tyre, though air leaves through the valve (contradicting the puncture).
And what exactly is hard to understand about the tyres having a tube? Isn't that rather standard for bicycle tyres?

The inner tube of the ST2 has schräder valves. These are also used for cars.
You need a bicycle pump that is compatible with this type of valve or an adapter to put on.
You need to attach the pump it firmly to the valve (press it down well), otherwise the air wil escape from the valve, just as you describe.
